How it works
RIP-seq uses a target-specific antibody to isolate RNA-protein complexes. The recovered RNA is sequenced to identify associated transcripts and compare their enrichment. Antibody selection, input samples and controls are considered together during study design.
Applications
- Identify coding and noncoding RNAs associated with the target protein
- Compare RNA enrichment across experimental conditions
- Investigate the roles of RNA-binding proteins in post-transcriptional regulation
Key features
- Transcriptome-wide screening of protein-associated RNAs
- Supports analysis of mRNA, lncRNA and other RNA classes
- Complements CLIP-seq with an enrichment-based view of associated transcripts
Workflow
- 01
Assess the samples, target RNA-binding protein and antibody
- 02
Immunoprecipitate RNA-protein complexes
- 03
Purify RNA, prepare libraries and sequence
- 04
Identify enriched RNAs and analyze their functions
