Epigenomics

ChIP-seq

Map transcription factor binding and histone modifications by sequencing DNA enriched with a target-specific antibody.

How it works

ChIP-seq combines chromatin immunoprecipitation with sequencing to identify genomic regions associated with a protein of interest. Target-specific antibodies enrich the relevant DNA fragments, enabling studies of histone modifications, transcription factor binding and nucleosome-associated regulation.

Experimental and analytical workflow

Applications

  • Map histone modifications and investigate their relationship with gene expression
  • Identify binding regions of transcription factors and other chromatin-associated proteins
  • Compare protein-DNA interactions across experimental conditions

Key features

  • Genome-wide identification of enriched regions
  • Antibody selection and sample requirements considered during project planning
  • Complements ATAC-seq, RNA-seq and Hi-C in studies of gene regulation

Workflow

  1. 01

Define the target protein, antibody and control design

  2. 02

    Prepare chromatin and perform immunoprecipitation

  3. 03

Purify DNA, construct libraries and sequence

  4. 04

Identify, annotate and compare enriched regions
