How it works
Hi-C captures chromatin contacts through crosslinking, digestion and proximity ligation, followed by sequencing to construct genome-wide interaction maps. These maps support comparisons of chromosome organization across conditions and integration with gene expression and epigenomic data.
Applications
- Compare chromatin contact patterns across experimental conditions
- Combine ChIP-seq with Hi-C to study histone modifications and genome organization
- Relate RNA-seq expression changes to features of the 3D genome
Key features
- Genome-wide profiling of spatial chromatin interactions
- Study design can include an in situ Hi-C workflow
- Sequencing depth and analysis selected for the required resolution and sample characteristics
Workflow
- 01
Assess samples and crosslink chromatin
- 02
Digest DNA, label fragments and perform proximity ligation
- 03
Enrich contact fragments, prepare libraries and sequence
- 04
Filter valid contacts and analyze contact matrices and genome organization
We tailor the experimental workflow and analysis to your samples and study objectives.
